Quantification of bleomycin induced lung injury by means of H magnetic resonance elastography

چکیده
Introduction: Interstitial lung disease (ILD) induced end-stage fibrosis is a multiphase process that includes: 1) an initial or exposure phase; 2) an exudative phase; and 3) a final stage with one of two outcomes; a) edema clearance associated with alveolar wound healing and recovery, or b) organization of the alveolar space filling material followed by scar formation associated with loss of normal structure and function. It has previously been demonstrated that an MR-based method of quantitating shear modulus (μ) known as magnetic resonance elastography (MRE) can be used to assess normal lung [1]. However, the ability to quantitate lung injury with MRE and whether or not these changes differ from normal lung remains unknown. The purpose of this work is to investigate whether or not MRE-based estimates of μ reflect changes to normal rat lung parenchyma following administration of bleomycin, a known agent for regulation of pulmonary fibrogenesis.

Saffron Protection against Bleomycin-Induced Pulmonary Fibrosis in Rats
Background: Bleomycin-induced lung fibrosis has been accepted as an animal model for fibrosis in rats. The aim of this study was to evaluate the effects of saffron aqueous extract on this disorder paving the way for more investigation in treating idiopathic pulmonary fibrosis in human. Methods: Male Wistar rats (250–300 gr) were instilled a single dose of bleomycin (5 mg/kg) via in...
متن کاملLung volume quantified by MRI reflects extracellular-matrix deposition and altered pulmonary function in bleomycin models of fibrosis: effects of SOM230.
Idiopathic pulmonary fibrosis is a progressive and lethal disease, characterized by loss of lung elasticity and alveolar surface area, secondary to alveolar epithelial cell injury, reactive inflammation, proliferation of fibroblasts, and deposition of extracellular matrix. The effects of oropharyngeal aspiration of bleomycin in Sprague-Dawley rats and C57BL/6 mice, as well as of intratracheal a...
متن کاملAdministration of Bleomycin via the Oropharyngeal Aspiration Route Leads to Sustained Lung Fibrosis in Mice and Rats as Quantified by UTE-MRI and Histology
Pulmonary fibrosis can be experimentally induced in small rodents by bleomycin. The antibiotic is usually administered via the intratracheal or intranasal routes. In the present study, we investigated the oropharyngeal aspiration of bleomycin as an alternative route for the induction of lung fibrosis in rats and mice. The development of lung injury was followed in vivo by ultrashort echo time m...
متن کاملCombined Effect of N-Acetyl Cysteine and Clarithromycin on Bleomycin Induced Pulmonary Fibrosis
Combined therapy with clarithromycin (CLTR) and N–acetyl cysteine (NAC) may be useful in diseases with impaired oxidant-antioxidant balance, fibroblast proliferation, and collagen deposition such as pulmonary fibrosis. Activated inflammatory cells which accumulate in the lower airways may release increased amounts of reactive oxygen species (ROS) when accompanied with a deficiency in glut...
متن کاملEffect of thalidomide on the alveolar epithelial cells in the lung fibrosis induced by bleomycin in mice
Introduction: Idiopathic pulmonary fibrosis (IPF) is a relentlessly progressive and usually fatal lung disease of unknown etiology for which no effective treatments currently exist. In the adults type I and II pneumocytes, forms Components of the alveolar epithelial cells. In this study, we investigated the effect of thalidomide on the alveolar epithelial cells (type I and II pneumocytes) in ...
